Yes, definitely. Just because an actor is extremely skilled does not necessarily mean he or she will be suited for the character the movie is trying to portray, just as how in music an extremely skilled rock singer is not necessarily suited to sing jazz if he or she has never understood or learned the genre, or how being a Physics graduate does not necessarily make one a decent engineer. It is not just skill, technique, or whatever you call it - it is how you sell the role and whether people buy to it or not.

One very good example of this is George Clooney in Batman and Robin (1997).

The man is a great actor and has an overall very good filmography, but he was highly miscast for this movie, because putting the bad looking suit as well as the bat nipples aside, Clooney lacked the grit, ‘badassery’, and darkness that the character is well-known for, unlike, for instance, Ben Affleck in Batman v Superman: Dawn Of Justice (2016), who despite Zack Snyder’s polarising direction for the character (i.e. Batman killed people in this film), simply nailed it to even the most minute of details.
